WHAT HAVE I LEARN?
This week we continue the experiment on basic laboratory techniques which is the techniques for Isolation of Pure cultures and also learn about cultural characteristics if organisms. Isolation technique is used to isolate discrete colonies initially require the number of organisms in the inoculum to be reduced. There are three method that we can use in isolation technique that is the streak plate method, spread plate technique and pour plate technique. I have learnt about the streak-plate technique and spread-plate technique.
I'm able to obtain single colony on agar plate. The type of microbes that isolate can be seen clearly as they have different growing characteristics. S. marcescens is pink in colour while S. aureus is yellowish white. Each type of microbes which are subculture in nutrient agar, broth and also gelatin can be differentiate between each other by their size, pigmentation, form of growth, margin and elevation. From the experiment, I found out that the M. lutues are slow growth which can't see much grow within a day. The P. aeruginosa grows at a high rate which perform in green colour.
