BMY3201-1 BASIC MICROBIOLOGY TECHNIQUES
WHAT HAVE I LEARN?
This week, we move on to part two which learn about microscopy. Microscopes have two main type which are light microscope and electron microscope. Light microscope including brightfield microscope, darkfield microscope, phase- contrast and fluorescent microscope. Light microscope use visible light or ultraviolet rays to illuminate specimens. For electron microscope, there are scanning electron microscope and transmission electron microscope which use electron beams and magnets to observe submicroscopic particles. We also learn about the parts of compound microscope and the way to handle it. Dr also teach us about what to do to clean the microscope after using it with oil immersion. We able to use see a few prepared microorganisms under the microscope under high power and oil immersion.
We learn on how to use a hanging drop preparation and wet mount for microscopic examination of living microorganisms. I failed to observe the microorganisms at the beginning, the demo told me that i put too little culture onto the depression slide, too much air inside that's why i can't observe anything. I try to do it again and finally i get to see the microorganisms under the microscope. I was so excited when I be able to see some movement of microorganisms.
